Hi Eric,
Mine were mostly long triathlons (like 8-10 hour events), but I did do one. First you'll need a fitness base in each of the events. Do the swim, the bike and the run at least 30 minutes each twice a week at a somewhat strenuous pace, for example for three weeks (if you haven't already).
Then, I would cut back to once a week on each and intersperse interval training on each one (see links on the top of my profile).
If your first triathlon is three or more months away, you can add weight training twice a week. If it's only a month away, you may have too much on your plate and in addition may not see significant benefit from the weights.
Listen to AJ and Others, too!

My general plan is to get to the gym 4 nights a week starting toward the end of this month (gym opens in a week). I am going to start lifting weights this winter and my cardio will be alternating between bike and treadmill. I'm also going to the local high school open swim and swim laps there when I can.
I am going to run a 5k with a friend in March, so that should tell me how I am doing with my fitness level. He wants me to run a 10k too, but we'll see about that. In the spring I can start putting 2 events back to back to see how my endurance is.
Thanks!
Eric
I just ran a sprint duathalon today. Toughest challenge yet. I came in last in my age group (8/8 50-60 year olds). You will need to train hard. I consider myself extremely fit and have done cycling events and 5-and-10k runs all year, but this one really took every effort to complete. It was a 1.5m run, 20m bike ride (headwinds and hills), and 3.1m run. It was also extremely windy and wet. I also was extremely intimidated by the "professional" athletes participating in this event. It seemed like everyone had a 4k bike and time trial alloy wheels and helmets.
I finished in 1hr 52m. The transitions are tough. Starting the last run my hamstrings were killing me and I had to run through the pain. I finished, and look forward to the next one but wow it's tough. I admire A.J.'s efforts even more now.
Good luck to you. Train hard and keep the goal in front of you.
